【摘要】:Biodeteriogenicmicrobesinfectingfuelsystemstypicallyaremostabundantwithinslimeaccumulationsonsystemsurfacesoratthefuel-waterinterface(GuideD6469).However,itisoftenimpracticaltoobtainsamplesfromtheselocationswithinfuelsystems.Althoughthenumbersofviablebacteriaandfungirecoveredfromfuel-phasesamplesarelikelytobeseveralordersofmagnitudesmallerthanthosefoundinwater-phasesamples,fuel-phaseorganismsareoftenthemostreadilyavailableindicatorsoffuelandfuelsystemmicrobialcontamination.GrowthMediumSelectivity8212;GuideE1326discussesthelimitationsofgrowthmediumselection.Anymediumselectedwillfavorcolonyformationbysomespeciesandsuppresscolonyformationbyothers.Asnotedin6.3,physical,chemicalandphysiologicalvariablescanaffectviablecellenumerationtestresults.TestMethodD7463providesanon-culturemeansofquantifyingmicrobialbiomassinfuelsandfuelassociatedwater.Sinceawiderangeofsamplesizes,ordilutionsthereof,canbeanalyzedbythemembranefiltertechnique(TestMethodsD5259andF1094),thetestsensitivitycanbeadjustedforthepopulationdensityexpectedinthesample.Enumerationdatashouldbeusedaspartofdiagnosticeffortsorroutineconditionmonitoringprograms.Enumerationdatashouldnotbeusedasfuelqualitycriteria.1.1Thispracticecoversamembranefilter(MF)procedureforthedetectionandenumerationofHeterotrophicbacteria(HPC)andfungiinliquidfuelswithkinematicviscositiesx2264;24mm2x00B7;s-1atambienttemperature.1.2ThisquantitativepracticeisdrawnlargelyfromIPMethod385andTestMethodD5259.1.3Thistestmaybeperformedeitherinthefieldorinthelaboratory.1.4Theabilityofindividualmicrobestoformcoloniesonspecificgrowthmediadependsonthetaxonomyandphysiologicalstateofthemicrobestobeenumerated,thechemistryofthegrowthmedium,andincubationconditions.Consequently,testresultsshouldnotbeinterpretedasabsolutevalues.Rathertheyshouldbeusedaspartofadiagnosticorconditionmonitoringeffortthatincludesothertestparameters,inaccordancewithGuideD6469.1.5Thispracticeoffersalternativeoptionsfordeliveringfuelsamplemicrobestothefiltermembrane,volumesordilutionsfiltered,growthmediausedtocultivatefuel-bornemicrobes,andincubationtemperatures.Thisflexibilityisofferedtofacilitatediagnosticefforts.Whenthispracticeisusedaspartofaconditionmonitoringprogram,asingleprocedureshouldbeusedconsistently.1.6ThevaluesstatedinSIunitsaretoberegardedasstandard.Nootherunitsofmeasurementareincludedinthisstandard.1.7Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
